In a total of 276 mothers of a child with a CHD in case-control family study, comprising of 190 outflow tract defects (OTD) and 86 non-outflow tract defects (non-OTD) and 324 control mothers of a non-malformed child, in mothers filled out general and food frequency questionnaires at 16 months after the index-pregnancy, maternal diet high in saturated fats and low in riboflavin and nicotinamide seems to contribute to CHD risk, in particular OTDs.

Dr. Smedts HP, the lead author said," mothers of a child with OTD, had higher dietary intakes of saturated fat, 30.9 vs. 29.8 g/d; P < 0.05. Dietary intakes of riboflavin and nicotinamide were lower in mothers of a child with an OTD than in controls (1.32 vs. 1.41 mg/d; P < 0.05 and 14.6 vs. 15.1 mg/d; P < 0.05, respectively)".

Although vitamin B2 plays an important role in reduced risk in developed congenital heart defects, other research suggested that diet low in vitamin B12 and imbalance in the maternal intake of proteins and low folate intake may also associate with an increased risk of a child with a CHD, according to the study of 192 mothers of a child with a CHD and 216 mothers of a healthy child. filling out food frequency questionnaires covering the current dietary intake.

Other study suggested that maternal diet high in saturated fats and low in riboflavin and nicotinamide seems to contribute to CHD risk, in particular OTDs.

Therefore, vitamin B2, may be associated to reduce risk of congenital heart defects, other nutrients factors should also be taken into account as well.

The association of thiamine deficiency among diabetes mellitus without treatment may increased risk of vascular disease involving blood vessels – the arteries and veins of the circulatory system.

According to the Queen Alexandra Hospital, thiamine deficiency may lead to hyperglycaemic-induced damage, affecting vascular disease in promoting cardiovascular risk.

In a study of diabetic patients (26 type 1 and 48 type 2) with and without microalbuminuria and 20 normal healthy control volunteers, researchers found that decrease plasma volumes are associated to patients with diabetes as shown in the scale. Dr. Thornalley PJ, the lead author said, "low plasma thiamine concentration is prevalent in patients with type 1 and type 2 diabetes, associated with increased thiamine clearance".

Taking altogether, low plasma thiamine concentration in diabetic patients showed to increased risk of vascular disease. Intake of high dose thiamine may be beneficiary to dysfunction of intracellular transport or enzymatic abnormalities in ameliorate the development of vascular dysfunction in inducing stroke.

In the recruited 50 patients with PD (33 men and 17 women; mean age, 70.4 ± 12.9 years; mean disease duration, 7.3 ± 6.7 years) with 100 mg of thiamine administered intramuscularly twice a week, without any change to personal therapy, thiamine group showed a significant improvement of symptoms of PD, reduced selective neural damage in the centers and provoking neurodegeneration.
And the re-evaluated after 1 month and then every 3 months during treatment showed a clinical stable improvement over time in all the patients.

Other study suggested that high dose of thiamine as initial treatment may attenuated motor and non-motor deficits of patients with PD.

A recruited three patients with newly diagnosed PD who are not involved in anti-Parkinson's therapy. with Plasmatic thiamine within healthy reference range study, therapy with high doses of thiamine showed a considerable improvement in the motor part of the Unified Parkinson's Disease Rating Scale(UPDRS) ranging from 31.3% to 77.3% which is considered as impotant levels in reduced risk of selective neuronal damage caused by thiamine deficiency due to a dysfunction of thiamine metabolism.

In support to the above analysis, researchers at the Vietnamese American Medical Research Foundation insisted that thiamine supplement can act through a number of non-genomic mechanisms that include protein expression, oxidative stress, inflammation which may link to development of PD.
Taking together, elevated thiamine levels might provide protection against PD, but using high doses can only be prescribed by profession medical doctors.

Diabetes is a condition caused by insufficient insulin entering the bloodstream in regulating the glucose. The disease is either caused by cells in pancreas dying off or receptor sites clogged up by fat and cholesterol. Thiamine therapy induced a significant change of protein functions in reduced symptoms and complications in patients with Type II diabetes, a study by renowned Institute suggested.

In a double-blind, randomised trial, where 12 hyperglycemic subjects (10 cases of impaired glucose tolerance and 2 new cases of type 2 diabetes mellitus) received both placebo and thiamine capsules (3 × 100 mg/day) for six weeks in a cross-over manner, high dosed vitamin group showed a significant effects on the blood pressure of individuals with hyperglycemia at early stages, and prevention of vascular complications.

Furthermore,  high-dose thiamine supplementation also displayed an impact on glucose tolerance in patients with impaired glucose metabolism.

Dr. Alaei Shahmiri F, the lead researcher on the study said, " supplementation with high-dose thiamine may prevent deterioration in fasting glucose and insulin, and improve glucose tolerance in patients with hyperglycemia. High-dose thiamine supplementation may prevent or slow the progression of hyperglycemia toward diabetes mellitus in individuals with impaired glucose regulation"

Further study also suggested that increase supplement intake may reduce risk thiamine deficiency in the diabetic population and ameliorate the early sign of diabetic kidney disease and improve lipid profile.

Taking altogether, intake of thiamine should have a potential effect in reduced risk of complications and treatment of Type II diabetic patients, pending to further studies.

In the total of 437 consecutive patients who underwent laparoscopic RYGB or laparoscopic adjustable gastric banding, 303 were included in the study, thiamine deficiency is found to associated with 15.5% of the participants with. male patients presented with greater mean preoperative thiamine levels (3.2 microg /dL) than female patients (2.4 microg/dL).

Taking altogether, intake of vitamin B1 in prescribed doses may reduce risk of thiamine deficiency complications in obese patients.

The joint study also emphasize that since the survival rate of of patients with an alcohol-related Wernicke-Korsakoff syndrome is poor, and treatment of alcohol abuse and early diagnosis of thiamine deficiency is a priority for improving clinical outcomes in prevention of of complications such as severe impairment of cognitive function and a craving for alcohol.

There is no doubt that vitamin B1 treatment in early diagnosis of Wernicke-Korsakoff syndrome in a priority in management of patients may be neccessary to reduced complications and motility.

Diabetes is a condition caused by insufficient insulin entering the bloodstream to regulate the glucose. Taking altogether, intake of oral supplement of vitamin B1 in patients with diabetes may have a positive effect in managing diabetic complications, but quantity control is necessary to prevent overdoses.

Vitamin A, a bi-polar molecule formed by bonds between carbon and hydrogen, is a fat soluble vitamin which can not be stored in the liver but it can be converted from beta-carotene, a powerful antioxidant. In a total of 63,443 women and 38,603 men were followed up, from 1984 until May 31, 2010, in the Nurses' Health Study and from 1986 until January 31, 2010, in the Health Professionals Follow-up Study suggested, higher intake of bioavailable lutein/zeaxanthin is associated with a long-term reduced risk of advanced AMD, as risk of advanced AMD is reduced by about 40% in both women and men.

In fact, risk of lung cancer is increased for smokers as the result of depletion of vitamin A in the body induced imbalance of protein expression in the retinoic acid receptor.
